This study presents the complete genome sequence of Cytobacillus firmus strain T8, which was obtained from the rhizosphere soil of pepper (Capsicum annuum L.). The genome of the strain consists of a single chromosome with a total size of 4,383,751 bp and the GC content of 42%.
Keywords: Cytobacillus firmus, complete genome, pepper, rhizosphere
In this study,
The genomic DNA of identified strain was extracted using DNA purification kit (Promega, USA) according to the manufacturer’s instructions. The quality and quantity of the extracted DNA was assessed using a Qubit Flex fluorometer (Thermo Fisher Scientific, USA) and a NanoDrop One microvolume UV-Vis spectrophotometer (Thermo Fisher Scientific). The library was prepared following the manufacturer’s guidelines using the SQKLSK109 ligation sequencing kit (Oxford Nanopore Technologies [ONT], UK) with the NEBNext companion module (New England Biolabs, USA). Sequencing was performed on the MinION platform (ONT) with a FLOMIN111 R10.3 flow cell for 48 hours at the KNU NGS Core Facility (Republic of Korea).
To generate FASTQ files, base calling was carried out using Guppy v4.4.1 software in high accuracy mode (HAC). For quality trimming, sequences with Phred scores of < 7 were excluded from subsequent analyses.
